Inside the cell, acetate groups are cleaved by intracellular esterases yielding a fluorescent molecule whose succinimidyl ester group covalently interacts with primary amines of intracellular proteins. CFSE is compatible with standard intracellular staining protocols using aldehyde fixation and saponin-based permeabilization.
